"E, adesso, devo andare al negozio"
"I have to" = "Devo" [ Voice of the verb "Dovere" ]
More over,
"E, adesso, vado al negozio" means "And now, I go to the store" or "And now, I'm going to the store".
"E adesso ho vado al negozio" means nothing.

Just a note.
"I have to" also has another translation in Italian, almost identical to the english expression:
"I have to" = "(Io) ho da"
But you can't always use it.
